PIPES is mainly used as a biological buffer and can be used in laboratory research and chemical production processes. 

Application of PIPES

With the development of genome sequencing technology, research on the sequence, structure, and function of plant genomes is rapidly advancing. Obtaining high purity, content, and completeness of plant genome DNA is the primary prerequisite for the application of genome sequencing technology. In order to better study and understand the biological functions of genes and RNA, people have developed some methods for extracting RNA from plant tissues. The traditional methods for extracting plant RNA mainly include Trizol method, SDS method, CTAB method, guanidine isothiocyanate method, etc. The above method mainly involves extracting after adding water-based buffering agents and organic extractants. Nucleic acids are retained in the aqueous phase, and many impurities are removed by leaving them in the organic phase. However, a small amount of nucleic acid pollutants will still be retained in the aqueous phase and need to be removed in further purification steps. Therefore, new alternative methods have emerged, mainly based on selectively adsorbing nucleic acids onto solid microcolumns, then removing pollutants through cleaning, and finally eluting purified nucleic acids from the microcolumns. However, as the steps of these nucleic acid extraction methods increase, the types of reagents used and the time required for nucleic acid extraction also increase. Therefore, the CN113717969A patent provides a reagent and method for quickly and inexpensively extracting nucleic acids from plant tissues. This method requires providing a type of lysis, binding, and washing reagent, which comprises at least one salt compound capable of dissolving plant nucleic acids, at least one metal ion chelating agent that prevents DNA degradation, at least one buffer compound, water-soluble organic matter, phenol or water saturated phenol, and at least one mixture of RNase A enzyme inhibitors.

The above-mentioned buffer solutions can be TRIS, TRICINE, BICINE, MOPS, HEPES, PIPES, CHES, MES, etc.

The plant tissue nucleic acid extraction reagent proposed in this patent does not use toxic reagents and is environmentally friendly for the plant nucleic acid extraction process, beneficial to the health of experimenters and reducing environmental damage.
