3,3’,5,5’-Tetramethylbenzidine (TMB) is a chemical compound with a molecular formula C16H20N2. This organic compound is widely utilized in biochemistry and molecular biology as a substrate for the detection of peroxidase activity. TMB undergoes colorimetric changes in the presence of peroxidase, resulting in the development of a blue color. This makes it a crucial component in various enzyme-linked immunosorbent assay (ELISA) and immunohistochemistry applications.    

Application    
TMB's distinctive color reaction is employed to visualize and quantify the presence of specific biological molecules, such as antigens or antibodies, in various assays. The color change occurs due to the oxidation of TMB by peroxidase, leading to the formation of a blue product. The intensity of the color is directly proportional to the amount of peroxidase activity, providing a sensitive and quantitative measure for detecting biological interactions.

A TMB stock solution can be prepared by dissolving 1 mg/ml in DMSO. The working solution is made by diluting 1 ml of the DMSO stock with 9 ml phosphate citrate buffer, pH 5.0, and then adding 2 ml of fresh 30%
hydrogen peroxide per 10 ml of substrate/buffer solution immediately prior to use. Stock solutions of TMB in DMSO can be frozen in 1–2 ml aliquots at –20 °C. Solutions appear to remain active for at least two years when stored in this manner. Any oxidation would be revealed as a color change in
the working solution.
