AMPPD vs. Luminol, who will be the king in luminescent immunoassay? 2018-01-08 16:34:52

Both AMPPD and luminol are redox-based luminescent detection reagents. In contrast, ancient reagent luminol has been developed for nearly a century and can detect more than 100 substances; however, fluorescent labeling reagent AMPPD has only developed in recent decades, the report is relatively less. In the future, who will be superior?


Luminol, the chemical name is 3-aminophthalimide, it mainly used in blood testing. In the experiment, solution of hydrogen peroxide and hydroxide base is generally used as activator, in which hydrogen peroxide is catalyzed to decompose by iron-containing compounds. Which in turn reacts with the oxygen generated by the decomposition of hydrogen peroxide to produce 3-aminophthalic acid in the excited state. When it returns to the ground state, it will give out blue light. Hemoglobin in blood contains iron, which can catalyze the decomposition of hydrogen peroxide.


AMPPD, chemical name 3-(2'-Spiroadamantane)-4-methoxy-4-(3''-phosphoryloxy)phenyl-1,2-dioxetane, is used in the super high sensitivity detection of phosphatase. AMPPD is enzymatically hydrolyzed by AP to produce a fairly stable AMP-D anion under alkaline conditions and a suitable buffer with a catalytic half-life of 2~30min, giving sustained light at a wavelength of 470nm. The intensity reached the peak at 15min, and remained relatively stable for 15~60min. the rate of light emission depends on the concentration of alkaline phosphatase.


So which one is better?



Chemiluminescence detection reagents are divided into two categories, one is direct chemiluminescence reagent, the other is indirect chemiluminescence system. Both AMPPD (CAS 122341-56-4) and luminol belong to the indirect luminescence system and catalyze the luminescence by enzyme. The difference is that luminol is the detection substrate of horseradish peroxidase, and AMPPD is detection system of alkaline phosphatase. From the detection performance, AMPPD has the highest detection sensitivity, stable detection results, good repeatability, long light-emitting time, less affected by the environment, it is better than luminol.


It is believed that with the development of the technology, AMPPD (Model No.: L0044) will also become a luminescent detection reagent which is as well known as Luminol. In fact, besides luminol and AMPPD, acridinium ester and APS-5 are also luminescent immunoassay, no matter which one will be the most widely used in the future, there is one thing will not be changed, which is they all contribute to the development of science.
