Colormetric Determination of Metallothionein (MT) Using DTNB


Metallothionein is a small metal binding protein rich in cysteine, which is a natural active substance with extremely important physiological functions.


A corlometric method for the determination of metallothionein (MT) was established using 5, 5’-dithio bis (2-nitrobenzoic acid) (DTNB) as the derivatizing agent and measuring the absorbance of the colored product TNBA at 412 nm.

Determination of glutathione in Lycium barbarum by 5,5'-dithiobis (2-nitrobenzoic acid) DTNB


Glutathione is a tripeptide with a high concentration distribution in biological cells, which contains cysteine residues, and has important biological activities. The thiol group of glutathione has reducing ability, DTNB reacts with sulfhydryl groups in a pH7 solution to form 3-carboxy-4-nitrothiophenol. which are yellow and have a strong absorption peak at a wavelength of 412 nm. The absorption intensity of the absorption peak at the maximum absorption wavelength in the excess DTNB solution has a good linear relationship with the glutathione concentration.Therefore, spectrophotometry and mass spectrometry were used to determine GSH in plant samples.


In addition to being a sensitive reagent for the determination of sulfhydryl groups in proteins and peptide tissues, DTNB is also used for the colorimetric determination of uric acid and cholesterol, the determination of chlorodimethyl arsenic, and organic Monitoring of phosphorus pesticide poisoning (Determination of cholinesterase), etc.
