Application of Tris|CAS:77-86-1| in eye drops 2021-12-23 14:39:21

Dry eye syndrome is caused by the long-term lack of sufficient lubrication and moisture on the ocular surface. The symptoms of dry eye vary from person to person, ranging from mild symptoms to persistent eye inflammation, severe inflammation, and scarring on the front surface of the eye. In order to alleviate the symptoms of dry eye, we often use eye drops. CN 112804990 A discloses a liposome eye drops formula, which is composed of liposomes. The liposomes are composed of non-hydrogenated phospholipids. The non-hydrogenated phospholipids contain linseed oil, vitamin A palmitate, and vitamin E poly Ethylene glycol succinate and water phase vitamin B12 and Pycnogenol. The specific preparation steps are as follows:

1. Preparation of liposomes carrying lipophilic substances

① Alcohol solvent is usually used to dissolve all lipophilic components, not hydrogenated phospholipids, linseed oil, vitamin A palmitate and vitamin E polyethylene glycol succinate vitamins;

② Remove the solvent in vacuum to obtain a dry powder;

③ Disperse the powder in a borate buffer and mix it with a mechanical mixer to obtain a solution of large liposomes, which can be defined as proliposomes;

④ Then extrude this large liposome solution under a high pressure of 600 to 1000 bar; repeat the high pressure extrusion procedure several times, between 4 and 7 times, to obtain liposomes with a size less than 200nm;

2. Preparation of an aqueous solution containing hydrophilic substances

① Dissolve Pycnogenol, Tris, Vitamin B12, hyaluronic acid and borate buffer in distilled water;

The concentration of the lipophilic substance in the first step and the concentration of the hydrophilic substance in the second step should be added in a 1:1 volume to obtain a solution of the required concentration;

② Obtain a clear red solution;

3. Prepare the final sterile eye drops

Add a certain amount of "X" liposome solution prepared in step 1 to the same amount of "X" water color solution prepared in step 2, to obtain a certain amount of "2X" final standard eye drops, and finally standard eye drops are sterilized by filtering on a 0.2 micron filter.

The presence of Pycnogenol in the preparation method enhances the antioxidant capacity of the external aqueous phase; the presence of vitamin E polyethylene glycol succinate enhances the antioxidant effect on the lipophilic phase of liposomes; There are vitamin E polyethylene glycol succinate in the liposome, and Pycnogenol and vitamin B12 outside the liposome, which have a protective effect on UVA/UVB rays (shielding effect).
