The chemiluminescence method for the detection of SARS-CoV-2 antibodies has important clinical value. 3-(2-Spiroadamantane)-4-methoxy-4-(3-phosphoryloxy)phenyl-1,2-dioxetane (AMPPD) is an ideal chemiluminescent substanceand it is an important component of SARS-CoV-2 antibody detection reagent.


3-(2-Spiroadamantane)-4-methoxy-4-(3-phosphoryloxy)phenyl-1,2-dioxetane,the common name is AMPPD , and its CAS number is 122341-56-4. AMPPD is the latest ultra-sensitive alkaline phosphatase substrate in the field of biochemistry, and it has a wide range of applications in biological detection technology.

There is a tetra-heterocyclic ring and a phosphate group in which the -O-O- bond is weak in the molecular structure of AMPPD. The phosphate group maintains the stability of the entire molecular structure. Under normal circumstances, this compound is very stable, and the solid AMPPD stored at 5 ° C hardly decomposes. When alkaline phosphatase catalyzes, AMPPD undergoes hydrolysis of the phosphate group to form an unstable intermediate. This intermediate then decomposes on its own, and the -O-O-quaternary ring releases a large amount of energy, excites the chemiluminescence reaction, and emits photons.

AMPPD has a decomposition half-life of 2 to 30 minutes and emits continuous light with a wavelength of 470 nm. Its intensity reaches a peak at 15 minutes, and the light intensity remains relatively stable within 15 to 60 minutes.

Ping Liu and others [1]used AMPPD as a chemiluminescence substrate solution for the novel coronavirus igm/igg magnetic particle chemiluminescence immunodetection kit. Alkaline phosphatase or horseradish peroxidase catalyzes the luminescence of AMPPD to determine the luminescence value (RLU) of each sample tube. The luminescence value of the sample is positively correlated with the concentration of the new coronavirus IgM/IgG antibody in it, so as to detect the new coronavirus IgM/IgG antibody in the human serum.


Lihui Wen [2] used AMPPD as a luminescent substrate in a new  detection method of coronavirus sars-cov-2 S protein.

The enzyme conjugate catalyzes the emission of photons from AMPPD, and the number of photons is proportional to the concentration of S protein in the sample. The SARS-CoV-2 recombinant S protein is diluted to be used as the calibration product of the detection system. The concentration of S protein in the sample is calculated by the calibration curve of concentration-relative luminescence value (RLU).

The preparation method of AMPPD is relatively complicated, and there are not many published methods for industrial production.  Yacoo Science has invented a method for preparing AMPPD with high safety, low production cost and easy industrial production, and can supply high-quality AMPPD stably.
