Tris——Biobuffer? Drug? Or Pharmaceutical intermediate? 2017-09-01 16:32:50

Tris (Tromethamine) is well known as a kind of biobuffers for many biochemical experiments. In fact, it is also widely used in the medical field: It is a drug for acidosis, and an important pharmaceutical intermediate for a variety of drugs.


Biobuffer



Tris is alkaline, which can be prepared to buffers from acidic to alkaline only with HCl. Tris HCl CAS:1185-53-1 has the following advantages:

  • The preparation method is simple, and it can form a variety of buffer systems, such as TE, TBE, TAE and so on;
  • It has less effects on the biochemical process, and does not react with calcium, magnesium ions and heavy metal ions;

But with the following disadvantages:

  • The pH value of Tris-HCl is greatly affected by the solution concentration and temperature;
  • It is easy to absorb CO2, so the buffer should be sealed after preparation.


Drug for Acidosis


As a drug, Tromethamine CAS 77-86-1 can be used for acute metabolic acidosis and acute respiratory acidosis. Because of sodium free, it is especially suitable for patients with heart failure, and the rescue of low-temperature anesthesia, shock and cardiac arrest. It also can be used to treat ischemic intracellular acidosis after organ transplantation.



Pharmaceutical intermediates


As an organic amine, tris (tromethamine) can form salts which is easily soluble in water with poorly soluble drugs having acidic groups. The representative drugs are dexketoprofen trometamol, fosfomycin tromethamine, carboprost tromethamine, ketorolac tromethamine.



  • Dextroantrofen tromethamine: It is suitable for short-term pain of trauma and postoperative;
  • Fosfomycin tromethamine: mainly used for the treatment of non-complex urinary tract infection;
  • Carboprost tromethamine: mainly used for treatment for postpartum hemorrhage;
  • Ketorolac tromethamine: It can be used to relieve pain of all symptoms.


Tris (tromethamine) is widely used in the biological field and medical field, whether used as biobuffer, drug, or pharmaceutical intermediate, Tris has played a huge role for the development of science and saving lives.
