What is tromethamine?

What is the function of tromethamine in eye drops?


(1) Buffer to adjust the pH value


The pH value of normal tear is 7.2~7.4, eyes can withstand the pH range of 5.0~9.0. The pH value of the eye drops will directly affect the irritation of the eye and the effect of the drug. When preparing the eye drops, it is necessary to add buffer to adjust the pH of the solution so that it is close to tears. Tromethamine is a pH regulator widely used in eye drops.


(2) Intermediate for active ingredient


As an organic amine, tromethamine can form water-soluble complex with poorly soluble drug containing acidic groups. The most common eye drop containing tromethamine is ketorolac tromethamine eye drops. Ketoacetic is a non-steroidal anti-inflammatory drug that can be used for injection. ketorolac tromethamine is soluble in water and stable in aqueous solution. It has the effects of antipyretic-analgesic and antiinflammatory, and is commonly used for inflammation and pain after ophthalmic surgery.



Is it safe to contain tromethamine in eye drops?


First, as a drug that can be used for acute metabolic acidosis and acute respiratory acidosis, tromethamine is available to the human body;

Secondly, we talk about the toxicity of any substance with the dose. As a buffer, the dosage is not much, and there will be no security risks with normal use;

Third, animal experiments and clinical studies have shown that ketorolac tromethamine eye drops can significantly reduce corneal sensitivity, with strong analgesic, anti-inflammatory effect, but have no significant adverse reactions.


Tromethamine (CAS 77-86-1)-containing eye drops are safe. However, if there is any discomfort, it is recommended to see a doctor, but not using eye drops by yourself. It can achieve the best therapeutic effect only with professional guidance.
