Use of guanidine hydrochloride in high selectivity fluorescent probe for detection of hydrazine 2019-05-31 13:59:27


guanidine hydrochloride is often used as an intermediate in medicines, pesticides, dyes and other organic compounds, it is also an important raw material for sulfonamides and folic acid; it can be used as a strong denaturants in the extraction of total RNA from cells. At the same time, guanidine hydrochloride is often used in various buffers for biological experiments. Below, we will introduce its application in a kind of fluorescent probe with reactive high selectivity detection of hydrazine. 


Hydrazine is an unstable and volatile colorless flammable liquid that plays an important role in medicine, pesticides, dyes, chemicals and aerospace. Hydrazine has carcinogenic, neurotoxic, visceral damage and other toxicities, easy to be absorbed by oral, skin, inhalation, etc., China's environmental protection law clearly stipulates the mass concentration of strontium, requires that the concentration of strontium in surface water and fishery water should not exceed 10.0μg/L. Therefore, the development of methods for rapid, simple, sensitive and selective detection of cesium is very important in the environmental and biological sciences.


The researchers provided a reactive, highly selective detection of hydrazine fluorescent probes that specifically react with hydrazine and emit purple fluorescence under ultraviolet light for the detection of hydrazine. The technical solution used is:
1. Nopinone and 2-pyridinecarboxaldehyde are subjected to aldol condensation to give 3-(2'-pyridylmethylene) nopinone:
2. Condensation of 3-(2'-pyridylmethylene) nopinone with guanidine hydrochloride to give 7,7-dimethyl-4-(2'-pyridyl)-4,5,6,7-Tetrahydro-6,8-bridged methylene quinazolin-2-amine;
3. The reaction of 7,7-Dimethyl-4-(2'-pyridyl)-4,5,6,7-tetrahydro-6,8-bridged methylenequinazolin-2-amine and chloroacetyl chloride gives 2-chloro-N-(7,7-dimethyl-4-(2'-pyridyl)-4,5,6,7-tetrahydro-6,8-bridged methylene quinazoline) Acetamide.

Compared with the prior process, the present invention product 2-chloro N-(7,7-dimethyl-4-(2'-pyridine)-4,5,6,7-tetrahydro-6,8-bridged methylenequinazoline)acetamide was prepared from the natural renewable resource β-pinene derivative nopinone, which selectively reacts with hydrazine and emits violet fluorescence upon irradiation with 365 nm ultraviolet light. It can be used as a specific fluorescent probe for detecting nopinone and has good practicability.
