Method for preparing MOPS with low cost and low pollution 2019-01-25 16:50:15

MOPS,3-(N-Morpholino)propanesulfonic acid, is an important biological buffer, the buffer range is 6.5~7.9, which is widely used in biological and chemical experiments, such as electron transfer and phosphorylation in chloroplast thin layer, culture of Streptomyces coelicolus and the production of cephalosporins, and electrolyte system component in isoelectric focusing electrophoresis (IEF) of two-dimensional gel electrophoresis. So the preparation of MOPS Buffer is very important.


Problems in existing preparation techniques

Existing technical routes typically require the use of an organic solvent to dissolve and provide a reaction atmosphere to ensure that the morpholine is on C3 of propanesulfonic acid. The use of a large amount of organic solvent not only increases the cost of production, but also the volatility of organic solvents, which adversely affects the health of the operator. How to recycle the used organic solvent is also a laborious and costly task. In addition, the use of organic solvents results in yields that are always around 70%, which in turn increases costs.


New preparation method
The researchers used morpholine and propane sultone as raw materials, mainly including the following steps:
(1) dissolving morpholine in water;
(2) dropwise adding propane sultone to the morpholine aqueous solution obtained in (1), and ring-opening addition of propane sulfonic acid at 25 °C to obtain an aqueous solution of 3-morpholinepropanesulfonic acid (reaction time is 4-10 hours) );
(3) After completion of the reaction, vacuum removal of water and most of the residual morpholine, cooling and crystallization are carried out to obtain a crude 3-morpholinepropanesulfonic acid;
(4) Recrystallization of crude product, suction filtration and centrifugal dehydration;
(5) The crystal obtained in the previous step was washed with absolute ethanol to obtain the final 3-morpholinepropanesulfonic acid.
The preparation method uses water as a solvent and does not require the use of organic solvent. For the operator, the factors that endanger the health of the body are reduced, and there is no trouble of recycling the organic solvent, which will not cause environmental pollution problems and reduce the cost. The final yield is above 85% and the purity is ≥99% (HPLC).
