The Difference between MOPS and MOPSO as Buffer 2019-08-30 10:55:02

3-(N-Morpholino)propanesulfonic acid is abbreviated as MOPS, is a zwitterionic biological buffer often used in biologica and biochemical research. Its useful pH range is from 6.5 to 7.9. And it is recommended for the researches as follows.

(1) For denaturing gel electrophoresis of RNA

(2) For protein purification in chromatography

(3) To measure adsorption during UV/VIS spectrophotometry and to study redox properties using cyclic voltammetry

(4) In nitrogenase electron transfer mechanism

(5) In the separation of nucleic acids and proteins by electrophoresis

(6) In the control of culture media pH, including cell culture media for yeast, bacteria and mammalian cells

(7) It has been used as a buffer component of charcoal yeast extract medium

(8) It interacts with peptide backbone of bovine serum albumin leading to a net stabilization of the protein


MOPSO is 3-Morpholino-2-hydroxypropanesulfonic acid. MOPSO is a zwetterionic aminosulfonate buffer that is very similar in structure to MOPS, differing by the presence of a hydroxyl group on C-2 of the propane moiety. MOPSO is belonged to Good buffers family which Good et al. developed to provide buffers in the pH range of 6.15-8.35 for wide applicability to biochemical studies.


MOPSO has been utilized as a buffer component in the analysis of copper by two distinct methods:

(1) A flow injection micellar technique of the catalytic reaction of the reaction bwtween 3-methyl-2-benzothiazolinone hydrazone with N-ethyl-N-(2-hydroxy-3-sulfopropyl)-3,5-dimethxoyaniline

(2) An electrospray-ionization quadrapole time-of-flight mass spectroscopy technique to measure the formation of chelating complexes of MOPSO with copper


As mentioned above, MOPS and MOPSO have different useful pH range. And they have respective advantages in the biochemical studies. So, buffer selection in the research depends on the studied system.
