C-reactive protein assay reagent with HEPES 2019-12-14 17:32:18

C-Reactive protein (CRP) is an acute phase reaction protein that reacts with S. pneumoniae C polysaccharide to form a complex with a molecular weight of 105,500 D and a half-life of 19 hours. Traditionally, CRP is a non-specific inflammatory marker, but nearly a decade of research has revealed that CRP is directly involved in cardiovascular diseases such as inflammation and atherosclerosis, and is the most powerful predictor and risk factor of cardiovascular disease. Therefore, the CRP content can be used as a diagnostic standard for both diseases. The reference range is as follows:

1. CRP is very low in healthy human serum (<3 mg/L), and its concentration is significantly increased in bacterial infection or tissue damage.

2. A CRP value of 10 to 50 mg/L indicates mild inflammation, such as local bacterial infections (such as cystitis, bronchitis, abscess), surgery and accidental trauma, myocardial infarction, deep vein thrombosis, inactive connective tissue disease, Many malignant tumors and most viral infections.

3. A CRP value of about 100 mg/L indicates a more serious disease, and its degree of inflammation requires intravenous injection if necessary.

4. A CRP value greater than 100 mg/L indicates a serious disease process and often indicates the presence of a bacterial infection.

However, the reagents in the prior art cannot cover the above reference range at the same time, which causes limitations in application and increases the cost of diagnostic analysis. Therefore, the developer developed a C-reactive protein assay reagent, including C-reactive protein detection reagent R1, C-reactive protein detection reagent R2 and C-reactive protein calibrator:

1. C-reactive protein detection reagent R1 components and content: 0.02~10% stabilizer, 10~100mM buffer, 0.02~0.05% preservative;

2. The components and contents of the C-reactive protein detecting reagent R2 are: 0.01 to 15% stabilizer, 1-3 mg/ml of C-reactive protein immunoglobulin microspheres, 10 to 100 mM buffer, 0.02 to 0.05 % preservative;

3. C-reactive protein calibrator, its components and content are: 0.01-13% calibrator stabilizer, 20mg/L C-reactive protein, 10-200mM buffer, 0.02~0.1% calibrator antiseptic Agent

The stabilizer is one of 0.6~1.5% sodium chloride and 0.1~1% BSA; the buffer is 4-hydroxyethylpiperazineethanesulfonic acid-sodium hydroxide buffer, trishydroxymethylaminomethane- Hydrochloric acid buffer, 3-(N-morpholinyl)-2-hydroxypropanesulfonic acid-sodium hydroxide buffer and phosphate buffer are prepared according to the mass ratio of 1:1:1:1; The protein-immunized latex microspheres are polystyrene microspheres; the preservative is one or both of proclin 300, gentamicin, and chloramphenicol.

The method uses 4-hydroxyethylpiperazineethanesulfonic acid-sodium hydroxide buffer, tris(hydroxymethylaminomethane-hydrochloric acid buffer), 3-(N-morpholinyl)-2-hydroxypropanesulfonic acid. - Sodium hydroxide buffer and phosphate buffer are prepared according to the mass ratio of 1:1:1:1, the effect is very remarkable; it has the advantages of high accuracy, high measurement accuracy and analytical sensitivity.
